Deputy chief of mission Bryan Hunt said the regional office will officially open by early October.
Hunt, who has been in the country for almost three weeks, said the office is an initiative of President Obama to increase aid assistance to Pacific nations.
The new office will look closely into mother-to-child HIV/AIDS transmission prevention as well as climate change in the region.
